Abstract

The use of methylation analysis in the structural elucidation of polysaccharides and glycoconjugates is discussed. Practical aspects of the permethylation reaction and the transformation into alditol acetates of the monomeric sugars obtained on acid hydrolysis is considered. Gas liquid chromatography and mass spectrometry of partially methylated alditol acetates are emphasised and a number of reference values for the former and reference spectra for the latter are given.
